Codine and morphine?

The analgesic action of codeine is about 20% as effective as morphine, however less addictive.

Is that enough evidence to conclude that the replacement of -OH groups with -OCH3 groups in this class of drugs will always change the properties in this way? Why or why not?

In a drug test a break down product of codien is morphine so how can that be?

    Codeine is considered a “prodrug”. It is metabolized in the body to the primary active compounds morphine and codeine-6-glucuronide. Roughly 5-10% of codeine will be converted to morphine, with the remainder either free or conjugated to codeine-6-glucuronide(~70%) or converted to norcodeine(~10%) and hydromorphone(~1%).

    That you have only looked at codeine vs. morphine is NOT enough conclusive evidence to make that broad of a statement. A more convincing argument could be made if several related hydroxy alkaloids were O-methylated and the bioactivity of the free hydroxy versus methoxy were measured.
